Example of a Transfection

(Transfection in the presence of 10% fetal bovine serum using NovaFECTOR)

1. Grow cells in 35mm dish using a medium
that contains 10% fetal bovine serum to
55-70% confluence.

2. Aspirate old medium from dish.

Add plasmid DNA and NovaFECTOR lipid to 1.0 ml of serum free medium (in 1.5 ml siliconized microtube). Let sit at room temperature for 15 minutes to allow DNA:lip complex formation. (Start with 2-3 ug of DNA and 8-10 ug of lipid, then change the concentration of DNA and/or lipid to optimize)

3. Pipet 1.0 ml of medium contains
20% fetal bovine serum to dish.

4. Add DNA:lipid complex to dish.

5. Incubate to express reporter gene product.
